研究概要

(1)我々が開発したin vitroのT細胞分化を解析するassay系(proc.Natl.Acad.Sci88;7548ー7551,1991)を用いてtoleranceの機構を解析した結果、胸腺の上皮細胞は、negative selectionには関与していないことが明らになった(manuscript in preparation)。
(2)インスリン非依存性糠尿病(NIDDM)のモデルマウスKKーAYマウスを用いて解析した結果、骨髄移植のみで、糠尿病が治療できるだけでなく、hynerinsulinemia,hypercholesteremla等の代謝異常が改善されることが明らかになった。さらに糠尿病性腎症も改善されること、また免疫学的異常としては、CD8^+T細胞の加齢による減少(末血中に)が認められるが、骨髓移植によりCD8^+T細胞が回復することが判明した。(J.Exp Med,in press)。
(3)慢性腎炎の中でも、ステロイドホルモン抵抗性の腎炎、focal glomerular sclerosis(FGS)のモデルマウスを用いて解析した結果、FGSも骨髄移植により治療できることが明らかになった。現在、このマウスの免疫学的異常を解析中である。さらにFGSの骨髄を正常マウスに移植することによって、FGSの病変が正常マウスにtransfer可能かを検討中である(manuscript in preparation)。
(4)新しいリウマチのモデルマウス、NZB/KNマウスに骨髄と骨(stromal cellsの補充を目的)を移植することにより、慢性関節リウマチの発症を予防できることを証明した(submitted for publication)。
(5)自己免疫疾患の新しいモデルマウス(NZW×BXSB)Flモデルマウスを用いて、胸腺と骨髄を正常マウスとの間で相互に移植し、このマウスの胸腺内のdouble positive T細胞の減少の原因が、胸腺の異常に起因するのではなく、骨髄の異常に起因することを見いだした(Manuscript in preparation)。
