胸腺細胞選択とT細胞抗原レセプター・CD3分子、アクセサリー分子の研究

研究概要	1.胸腺細胞4つのメジャーポピュレーションのうち、CD4^-8^-とCD4^+8^-細胞中にNK1.1抗原を発現するTCR α β^+亜群の存在を同定した。これらは加齢と共に増加し、全胸腺細胞の約1%にあたることが判明した。 2.NK1.1^+TCRαβ^+細胞は、Vβusageなど種々の点で通常のT細胞と異なる性状を示した。 3.NK1.1^+TCRαβ^+細胞はIL-2レセプターβ鎖を発現し、IL-2存在下で培養するとLAK活性を獲得した。 4.NK1.1^+TCRαβ^+細胞は、フレッシュな状態でも16時間のキラーアッセイにより同系、異系のCD4^+8^+末熟型胸腺細胞を殺すことが判明した。この細胞障害には、Fas抗原とFasリガンドの相互反応が必須であった。 5.lpr,gldなどの自己免疫マウスでは、NK1.1^+TCRαβ^+細胞の細胞障害活性は、ほぼ完全に抑制されていた。 6.胸腺細胞の分化に必須の骨髄由来単球・マクロファージ系細胞に発現する、サークファミリーのFgrに特異的な2H2単クロン抗体を樹立し、これを用いFgr発現を組織レベルで同定した。さらにFgrが、Ly6Cおよび70kDaの未知の蛋白と会合していることを証明した。 7.Csk遺伝子をトランスフェクトしたマクロファージ系細胞株を用い、Cskが種々のマクロファジー機能を抑制すること、しかし、Cskのover-expressionによって、これらの細胞がHgCl_2による細胞死からレスキューされることを明らかにした。 8.調べた範囲のマウスのI-A分子に結合して、免疫原性を示すペプチドのアミノ酸モチーフを決定した。これらを用いて、胸腺に発現するI-Aとペプチド分子の結合力が、胸腺細胞のpositive selectionにどのような影響を与えるか、研究継続中である。
