新しい免疫電顕法を用いた基底膜抗原蛋白が皮膚水疱症の発生に果たす役割についての研究

研究概要	本年度私達は昨年の結果を基盤に,自己免疫性水疱症である類天疱瘡患者血清中の抗皮膚基底膜自己抗体が認識する抗原についての研究を主体に行ったimmunoblotや分子生物学的手法を用いた研究により、類天疱瘡抗原などいくつかの基底膜抗原タンパクの生化学的特徴、あるいはcDNAシークエンスを解明してきた。しかしこのように生化学的にはかなり研究が進んでいるこれら基底膜抗原タンパクも、正常あるいは患者皮膚基底膜におけるその正確な局在部位に関しては,種々の困難な免疫電顕における技術的問題点のために未だ十分解明されていなかった。この理由は、従来まで皮膚免疫電顕の研究で主に使用されてきたペルオキシダーゼ標識抗体を用いたpreembedding免疫電顕法では、皮膚の微細構造の保持が十分できず、また巨大なペルオキシダーゼ反応産物が沈着部位の微細構造までもマスクしてしまうなどの要因によるものであった。今回私共は“全く化学固定を用いず、凍結固定・凍結置換のみで組織を超低温樹脂に包理して行うpostembedding免疫電顕法"に金コロイドをマーカーとして応用した。その結果、230kD類天疱瘡抗原はヘミデスモゾームの細胞内のみに、170kD類天疱瘡抗原はヘミデスモゾームの細胞質膜にそって細胞の内外にわたり、それぞれ別の部位に局在することを解明し得た(J Clin Invest:in Press)。また,イメージアサライザーを併用することにより、これら基底膜抗原の局在をより正確に、しかも定量的に解析する方法を開発することができた(J Histochem Cytochem 41:123-128,1993)。本研究の成果は、今後の皮膚水疱症の発生機序の解明に大きな貢献をし得たものと考えられる。
