研究概要	1.エイズウイルスコアタンパク質(p17^<gg>)のアミノ末端ミリストイルグリシンに対する特異抗体の調製ここでは便宜上エイズの原因ウイルス(HIVー1)をエイズウイルスと呼ぶ。ミリストイルグリシン(NーMyrーGly)を水溶性活性エステル化剤を用い、キ-ホ-ルリンペットヘモシアニン(KLH)及びウシ血清アルブミン(BSA)に結合させ、NーMyrーGlyーKLHを免疫抗原及びNーMyrーGlyーBSAをassay抗原として用いた。まず免疫抗原(10μg/0.1ml)をウサギに一週間毎に4回皮下注射し、1ヵ月後同抗原を静注し、4〜5日目に採血した。血清中のNーMyrーGly分子に対する抗体価をNーMyrーGlyーBSAを抗原として、ELISA法により求めた。 2.抗NーMyrーGly抗体の部分精製と抗体の特異性ウサギを常法により免役して得られたNーMyrーGlyに対する抗血清をNーMyrーGlyーBSA分子をアガロ-スゲルに固定化した抗原カラムを用いて部分精製した。抗体カラムに抗原抗体反応により吸着する抗体の量は血清1ml当り約1mgであった。この抗体の特異性を調べるため、脂肪酸分子の2メチレン鎖短いNーラウロイルーグリシン(NーLauーGly)ーBSA、2メチレン鎖長いNーパルミトイルーグリシン(NーPalーGly)ーBSA及び種々のグリシンペプチドを合成した。本抗体はNーMyrーGlyーBSA、NーLauーGlyーBSA、及びNーPalーGlyーBSAに反応し、BSA及び他のグリシンペプチドとは反応しなかった。この事から本抗体はNーfatty acylーGlyー分子を認識している事が明らかになった。 3.抗NーMyrーGly抗体によるエイズウイルスコアタンパク質の検出上記のように調製した抗体とエイズウイルスコアタンパク質NーMyrーGly p17^<gg>タンパク質との反応をウエスタンイムノブロット法及び ^<125>Iー放射免疫沈降法を用いて調べた。その結果、本抗体はp17^<gg>タンパク質と反応することが明らかになったが、p17^<gg>タンパク質以外にも数種のタンパク質と反応することが判明した。現在、更に本抗体の特異性を向上させるためNーMyrーGly分子に対する単クロ-ン抗体を調製中であり、次年度本単クロ-ン抗体の調製に全力を注ぐ予定である。
