Na^+ポンプにおける真時間での構造変化と分子内のカチオン移動

研究概要

Na^+,K^+-ATPaseをBIPMとFITCとで2重標識した標品を用いて、リン酸化反応中間体(EP)の形成と分解(モールド研究室)及び蛍光強度変化(ビーチャム研究室)を追跡した。BIPMからFITCプローブへの蛍光エネルギー移動はNa^+結合酵素の状態を基準にとると、Na^+閉塞酵素で減少し、酢酸感受性リン酸化酵素(E_1P)で増加、K^+感受性リン酸化酵素(E_2P)で最大に達し、K^+結合酵素で減少、Na^+結合酵素基準値に戻ることが示された。本酵素標品を用い、ポリクロメーターによる各反応中間体と分解のプロセスの測定を行った。Na^+閉塞酵素にアセチルリン酸(Acp)を添加すると、BIPM蛍光励起によるBIPM蛍光の増加(5/s)とBIPMプローブへからFITCプローブのエネルギー移動によるFITC蛍光の増加(11/s)と減少(0.7/s)が観察された。FITCプローブの単独励起でも増加(5/s)と減少(0.05/s)が観察された。BIPM蛍光は増加と共にred shift、エネルギー移動に基づくFITC蛍光はわずかにblue shift、直接励起のFITC蛍光はred shiftした。両プローブ間距離をForsterの式を用いて計算した。その結果Cys-964に結合しているBIPMプローブからLys-510に結合しているFITCプローブ距離は約36Aと推定された。各反応中間体形成と分解に伴うプローブ間距離の変化は約1A以内と推定された。現在この距離の算定に用いて配向性因子の値が各中間体形成と分解に伴って変化するか否かを検討中である。
H^+,K^+-ATPaseをBIPMプローブで修飾すると反応中間体形成に伴い、Trp残基とは相を異にするBIPM蛍光の変化が観察された。又EP形成に伴いTrp残基からBIPMプローブへの蛍光エネルギー移動の増加が示された。BIPM結合部位の同定が現在進行中である。
H^+ポンプへのATP結合とAcpからのEP形成量及びATPからのEP形成量が2:2:1であることが示された。これはH^+ポンプが2量体でATPを加水分解していることを示している。
