活性化T細胞におけるシグナル伝達とIL-3とGM-CSF遺伝子の発現機構の解析

研究概要	ヒトIL-3遺伝子のCT/GC rich regionはHTLV-I Taxに応答するエレメントである。このエレメントに結合するDNA結合蛋白質としてEGR1,EGR2に加えてZincフィンガーモチーフを持つ新規クローンDB1を単離、同定した。DB1は他の転写因子にもよく見い出される6個のZincフィンガーモチーフとグルタミンストレッチを持つ。特異的な抗体を用いたゲルシフトアッセイにより、DB1は構成的に、EGR1はT細胞活性化依存的にCT/GC rich regionに結合していることが明らかとなった。これらのZincフィンガー蛋白質をJurkat細胞に強制発現させることにより、IL-3プロモーターを上流配列および刺激依存的に活性化することが明らかとなった。さらに、マウス胸線腫細胞株EL-4を用いて、IL-5遺伝子発現はPKCとPKAの両シグナルにより活性化されるが、逆にPMA依存的IL-2遺伝子発現はPKAによって抑制されることが明らかとなった。ルシフェラーゼ遺伝子をレポーターとした一過性発現系を確立し、PKCとPKAに反応するIL-5遺伝子領域を同定し、NF-AT様配列(-117〜-92)、GATA様配列およびCLE0(-84〜-37)などの配列がIL-5遺伝子発現に必須であることを明らかにした。さらに、IL-2遺伝子領域においてPKAによる抑制を受けるエレメントの同定を試み、NF-AT,NF-κB,Oct配列などの複数のエレメントがPKAによる抑制に必要であることが明らかとなった。これらの結果から、cAMPに対する感受性の相違がTh1とTh2細胞のサイトカイン産生パターンを規定する一因である可能性が示唆された。PKAがIL-5とIL-2遺伝子発現に対して相反的な効果を示すが、その標的となっているシグナル伝達分子を同定することが必要である。
