トロンボモジュリンの抗凝固作用とエンドサイトシスにおける構造・機能相関の解析

研究概要	トロンボモジュリン(TM)は、血管内皮細胞上のトロンビンレセプタ-で、トロンビンによる血漿抗凝固因子のプロテインC(PC)の活性化促進因子である。TMはまた、トロンビンの凝固促進活性を直接的に阻害する作用もあり、血管内壁上の最も重要な凝固制御因子である。他方、内皮細胞上のTMは、TNFなどのサイトカイン刺激を受けて、細胞表面から消失するが、この理由の1つにエンドサイトシスが考えられている。本研究では、遺伝子工学的、蛋白工学的手法を用いて、TM上のトロンビンおよびPC結合部位の構造を明らかにするとともに、TMのエンドサイトシスに関係するTM分子内の特異的構造を明らかにすることを目的とする。本年度、申請者は次の2点を明らかにした。(1)TM上のトロンビン結合部位の解析:トロンビンはTMの細胞外領域に存在する6個のEGF(上皮成長因子)ドメインのいずれかに結合することが、これまでに明らかにされている。そこで今年度は、これらのEGFドメインのうちのどのドメインにトロンビンが結合するかを明らかにするため、複数のEGFドメインからなるリコンビナント変異蛋白質(例えば、EGF1〜6、EGF45、EGF56蛋白質など)を作製して検討した。その結果、トロンビンはTMのNH_2末端から5番目のEGFドメインに結合することが明らかになった。(2)TM上のPC結合部位の解析:PCはCa^<2+>を介してTMと相互作用し、主にリコンビナントTM蛋白質を用いた研究から第4EGFドメイン内にPC結合部位の存在が示唆されていた。そこで、TMの第4EGFドメインのAsp(349)をAlaあるいはGluに置換した変異TM蛋白質を作製して検討したところ、Asp→Ala変異でPC結合能は特異的に消失した。この結果からTM上のPC結合部位はAsp(349)と決定した。
