色素性乾皮症D群の相補遺伝子のクローニング

研究概要	色素性乾皮症(XP)のD相補群(XPD)は、XPAやXPCとともに、世界的に頻度が高く、高発癌性を示す。XPD群細胞はXPAに次いでUVによるピリミジン2量体や(6-4)光産物のDNA損傷の修復能を80-90%欠損する。まず大切なことはXPD細胞のUV感受性とDNA除去修復能の欠損を相補するcDNA遺伝子をクローニングすることである。これはXPDタンパクの分子修機構上での役割を知るためにも大切である。本年度は、pSVOriで不死化した日本人XPD患者由来のXP59TOpSV細胞にヒト正常cDNAのpcD2ライブラリー(G418耐性の選択マーカーであるneo遺伝子を発現する)の導入とG418/UV選択によって得られた21クローンにつき、UV生存曲線と不定期DNA合成(UDS)を検定した。その中もっともUV抵抗性であったpcD2-D17クローンでも平均致死線量Do=1.2J/m^<2、>UDS=35-50%であり、いずれも親株XP59TOpSV(Do=0.7J/m^2、UDS=40%)を十分に相補する結果が得られなかった。したがって、pcD2-cDNAの中に完全長のXPDcDNAがないことが示唆さされた。そこで正常cDNAをLTRプロモター下流に一方向性に挿入し、かつneo遺伝子をもつpCEV-cDNAライブラリー(10μg/ml)を大スケール(4回、総数5x10^7)でXPD群のXP59TOpSV細胞に導入し、選択した。究極的に2個の安全な部分的UV抵抗性のクローン(pCEV-D2,pCEV-D4)が得られた。これら2クローンは、Do=1.5-2.0J/m^2を示し、親株XP59TOpSV(Do=0.7J/m^2)の2-3倍の大きいDo値を示し、正常の1/2-1/3の部分相補が得られた。これらのクローンのUDSは正常の70-80%まで回復し、親株の2-3倍の回復を示した。より多くの修復能を獲得したpCEV-D2のゲノムDNAは約9kbのMluIのplasmid制限酵素断片をもち、約2.5kbcDNAを含んでいた。これは、ERCC2cDNAと同じNotI部位とPCR増幅を示すので、XPDcDNAが発現クローニングされたことを示している。日本人の3人XPD細胞のmRNAをRT-PCRすると、少なくとも1例にmRNA量の異常が見られた。
