ヒト正常2倍体細胞及びマウス脳における増殖停止誘導遺伝子の単離

研究概要	ヒト細胞における老化遺伝子の解析を3年間色々な新しい手法を考察しながら行ってきた。特にアンチセンスライブラリー法による不死化遺伝の単離の試み、ゲル内変性再会合を用いた組換え遺伝子の探索などは有望であったが、方法にまだ検討すべき点があり、満足な成果が得られていない。しかし、正攻法による老化遺伝子同定の研究は著しい進展を遂げたので、簡単に紹介する。細胞老化を終末分化あるいは癌抑制効果と区別するために、世界に2例しかない正常線維芽細胞由来の不死化細胞株KMST-6及びSUSM-1に着目した(それぞれγ線及び4NQO処理により分離された)。まず細胞の不死化は老化遺伝子の劣性変異によって生じることが示唆されているので、上記2株及びそれらの親細胞の全染色体についてRFLP解析を行い、意外にも7番染色体長腕の同じ領域にloss of heterozygosity(KMST-6)及び欠失(SUSM-1)を見出した。そこで、微少核細胞融合法を用いて、7番を含めたいくつかの染色体を上記2株及び癌細胞由来の対照群へ導入したところ、7番染色体だけが上記2株の細胞分裂を停止し、老化細胞に特徴的な形状を示した。7番染色体上の欠失は数Mbp以下(顕微鏡下では正常)と推定されるので、position libraryの作成及び7番染色体のradiation reduced hybridの解析によって、老化遺伝子が実際に欠失領域に存在するかどうか確認中である。以上から、正常細胞の老化を導く遺伝子が7番染色体上に存在することが初めて示され、それのクローン化の体制が築かれた。
