色受容細胞における分子応答

研究概要

交付申請書に記載の研究実施計画に沿って研究を進め、以下の知見を得た。1.アイオドプシンには塩素イオンが結合し、その結合によってアイオドプシンの吸収極大が変化する。そこで、塩素イオンの結合サイトを同定するために、塩素イオンを結合したアイオドプシン及び結合していないアイオドプシンとそれらの初期中間体(バソ中間体)との差FTIR(フ-リエ変換赤外)スペクトルを測定した。その結果、塩素イオンは発色団の14番目の炭素原子の近くのアミノ酸残基に結合していることがわかった。2.ConAーセファロ-スカラムとCMーセフイロ-スカラムを用いて、ニワトリ網膜から緑色感受性錐体の光受容蛋白質を分離・精製し、その蛋白化学的・分光学的性質をアイオドプシン及びロドプシンと比較した。その結果、錐体光受容蛋白質の性質として、非常に速く再生すること、また、メタIII中間体の存在しないことなどが明らかになった。3.遺伝子操作技術を利用してアイオドプシン以外の錐体光受容蛋白質(3種類)の一次構造を決定した。また、それらのアミノ酸配列を、すでに報告されているロドプシンと比較し、分子系統樹を作製した。その結果、光受容蛋白質はまず4種類の錐体光受容蛋白質に分岐し、その後錐体光受容蛋白質の一つからロドプシンが分岐したことが明らかになった。また、ロドプシン遺伝子を培積容養細胞に挿入して、ロドプシンの蛋白質を発現させることに成功した。4.錐体にG蛋白質トランスデュ-シンのγサブユニット(Tγ)の一次構造を明らかにするため、ウシ桿体Tγの全長cDNAをプロ-ブとしてニワトリ網膜のcDNAライブラリ-をスクリ-ニングしたが,Tγをコ-ドすると思われるcDNAクロ-ンは単離できなかった。一方、桿体TγのC末端システインの翻訳後修飾は、ロドプシンとG蛋白質との共役に必須であることを明らかにした。
