研究概要

交付申請書に記載の研究実施計画に沿って研究を進め、以下の知見を得た。
1.我々の研究から、低温で生成したアイオドプシンの中間体は、試料の温度を上げるともとのアイオドプシンに戻ることがわかっている。そこで、この反応がアイオドプシンに塩素イオンが結合していることに起因するかどうかを検討するために、塩素イオンを含まないアイオドプシン試料を調製して検討した。その結果、バソ中間体のアイオドプシンへの戻り反応は塩素イオンの有無によらないことがわかった。一方、メタI中間体からアイオドプシンへの戻り反応は塩素イオン特異的であり、そのKd値は約30mMと求められた。後者の結果から、アイオドプシンには塩素イオン特異的な光シグナル伝達の調節機構があることが示唆された。
2.ロドプシンのメタI中間体とルミ中間体の間に温度依存性の平衡があることを発見した。その結果、低温と室温でのロドプシンの光反応過程の違いが中間体の熱力学的パラメーターの温度依存性によって説明できる可能性が示唆された。
3.錐体型光レセプターであるオオヤモリのP467を培養細胞系で発現させることに成功した。発現させた蛋白質の吸収スペクトルは網膜から抽出したものと同じであることがわかった。また、ロドプシンのメタII中間体の挙動がC末端付近のシステイン残基の修飾(パルミチル化)の有無によって変化するかどうかを検討するために、部位特異的変異によってこのシステインを持たない変異ロドプシンを培養細胞系で発現させ、その光反応を検討した。その結果、バルミチル化の有無は、メタII中間体の挙動に影響しないことがわかった。
4.オオヤモリの光レセプターP521およびP467を網膜から分離・精製し、その分光学的な性質がロドプシンあるいはアイオドプシンのどちらに似ているかを検討した。その結果、両者から生成するメタII中間体の性質は、いずれもアイオドプシンに似ていることがわかった。
