黄色植物光捕捉顆粒の分子構築とカロテノイド・クロロフィル間エネルギー転移機構

研究概要

1.光捕捉系の単位構造を解析し、褐藻の場合は分子量約2万の蛋白一つにfucoxanthin 4,chlorophyll(以下Chl)a 4,Chl c_2 1分子が結合した複合体(FCPC)で、渦鞭毛藻(peridinin型)のそれは分子量約3万の蛋白一つにperidinin6-10,Chl a 2分子が結合した複合体(peridinin-Chl蛋白)で、Chl cを含まないことを明らかにした。
2.数種の褐藻で光捕捉系の大きさを閃光法で測定すると、反応中心あたりFCPCの数は30-90分子で、光照度の低いところで生育する褐藻ほど大きな値が得られた。これらの褐藻の光捕捉顆粒の大きさは、閃光法で求めた光捕捉系の大きさとほぼ一致し、光捕捉蛋白がこのような分子集合体の形で光捕捉に働いていることを示唆する。
3.褐藻と渦鞭毛藻の光捕捉蛋白の一次構造分析の結果、両者の間には全く類似がなく、血清学的にも相互の間に反応性は見られず、また、緑色植物のLHCPとも類縁性の低いことを明らかにした。
4.Diethyletherの処理で褐藻のFCPCからfucoxanthinだけが選択的に除去されることを見いだした。この処理をした蛋白にはChl cとChl aが除去されずに残るが、この両者の間のエネルギー転移はきわめて低い。しかしこの蛋白をfucoxanthinとincubateすることでもとの蛋白に近い吸収スペクトルをもつ標品が再構成され、この再構成された蛋白ではChl cからChl aへのエネルギー転移は回復し、また再結合したfucoxanthinにも高いエネルギー転移活性が見られた。FCPCに結合した状態のfucoxanthinの吸収は通常の溶媒中のものより長波長に移行しており、強い円二色性のシグナルを示すが、この状態は不安定で、容易に短波長型に移行し同時にエネルギー転移を行なわなくなる。再構成させた系でfucoxanthinが長波長型をとることは、蛋白との結合する環境がfucoxanthinの電子状態を修飾していることを示している。その一方でfucoxanthinの結合がChl cのエネルギー転移にかかわるような形で蛋白の構造を修飾することを示している。
