ス-パ-バイオリアクタ-の開発:有機溶媒中での高性能・特異的反応プロセスの構築

研究概要	水を基質の一つとする加水分解反応において、水の量が少ない場合には加水分解酵素が逆反応である合成反応を触媒した。脂溶性ラセミ化合物の光学分割を目的としたリパ-ゼによる立体選択的なエステル合成反応を利用して生物活性物質やその中間体となる光学活性有用化合物(キラルシントン)の合成を行った結果、世界に先駆けていくつかの基礎的な知見を得た。50種類以上のリパ-ゼおよびエステラ-ゼの有機溶媒中での活性について検討したところ、多くの酵素が一級テルペンアルコ-ルのシトロネロ-ルをエステル化しうることが明らかとなった。しかしながら、これらの酵素は一級アルコ-ルに対し立体選択性を示さなかった。また三級テルペンアルコ-ルはほとんど反応しなかった。メント-ルやボルネオ-ルのような二級アルコ-ルは、酵母リパ-ゼなどにより効率よく光学分割することができた。この際、溶媒としてはやはり低極性のものが良く、アシル供与体の種類は反応速度のみならず、反応の立体選択性にも大きく影響していた。すなわち、長鎖の脂肪酸はエステル化の基質としては極めて良好ではあるが、いずれのエナンチオマ-とも反応し、短鎖の脂肪酸では高い立体選択性がみられるものの反応速度は小さくなった。一方、2-(4-クロロフェノキシ)プロピオン酸のエステル化においては、アシル受容体である短鎖のアルコ-ルは大きな反応速度と低い立体選択性を、長鎖アルコ-ルはその逆を示した。固定化酵母リパ-ゼを用いてこのプロピオン酸誘導体の連続的なエステル化反応を行った場合、固定化酵素は4ヶ月以上高い活性と立体選択性を保持していた。さらに、このアシル受容体としてトリメチルシリルメタノ-ルの使用は反応性・立体選択性ともに極めて高い値を与えることがわかったが、これは炭素とケイ素の原子半径や電気陰性度などの違いによって説明することができた。
