軽度低脳温法による中枢ニューロン死防禦

研究概要	昨年度、我々は軽微低脳温が虚血侵襲に対して顕著なニューロン保護作用を発揮することを証明した。しかし、ニューロンが組織としての統合性のもとに機能している実験系を用いたので、上記の機構を解明するには余りにも複雑である。そこで今年度は、(1)より単純な系として培養細胞を用いての分析(2)低脳温化剤MK-801の薬効分析(3)軽微低脳温の臨床応用を考える際に問題とされる血液粘度への影響の三点について研究を行った。 (1)培養系での実験ラットの胎仔脊髄ニューロンの初代培養を用いた。200μMグルタミン酸に15分間暴露したのち、培養液中に放出される乳酸脱水素酵素活性を経時的に分析することにより、グルタミン酸毒性による細胞死の測定を行った。温度37℃と32℃の間に細胞死の程度について有意の差を認めず、低脳温の効果はインテグレートされた系でしか発揮されないと結論された。 (2)MK-801の薬効NMDA型グルタミン酸受容体チャネル阻害剤MK-801は、その脳温低下作用により虚血に対するニューロン保護効果を発揮する。その機構を解明するため、脳温をモニターしながら検索したところ、この物質は体温調節のセットポイントを見失い脳温は外気温の変化につれて動く変温動物様の状態になることが明らかとなった。 (3)イヌ血液粘度に対する温度の影響深麻酔下、イヌの総頚動脈バイパス法により、低脳温状態とした。外頚静脈より採取した血液の粘度をcone-plate型粘度計を用いて測定したところ、shear rate21secにおいて37℃より4℃低下で数パーセントのCP値の上昇(Hematocrit:40)が得られ、ヒト血液と同等の変化であった。
