研究概要	1)八木記念パ-クの近藤はヘテロでは白斑と毛の淡色化をおこし、ホモでは白色黒眼となるラットのミュ-タントを発見した。この表現型はマウスのW遺伝子座とSl遺伝子座のミュ-タントに酷似するため、ラットのミュ-タントWsがマウスのWに相当するのかSlに相当するのかをしらべた。Ws/Wsラットの骨髄細胞はWミュ-タントマウスの骨髄細胞と同様、放射線照射をうけたレシピエントの脾で増殖することができず、マスト細胞を欠損するWs/Wsラットの皮膚をヌ-ドラットに移植すると移植皮膚片中にマスト細胞が出現する。この移植実験の結果は、Ws/Wsラットのマスト細胞欠損を含む造血障害が、Wミュ-タントマウスの場合と同様、造血細胞の側にあることを示した。そこでWsミュ-タント遺伝子をより一層くわしく性格ずけるため、W遺伝子がすでにc-kit遺伝子と同一であることがわかっていたので、Ws/Wsラットと正常ラットからc-kitのcDNAをとり、その塩基配列をくらべた。Ws/Wsラットではc-kitcDNAのタイロシンカイネ-スをコドする領域に12塩基の欠損をみとめた。この12塩基がコ-ドナス4個のアミノ酸はいくつかのレセプタ-・タイロシンカイネ-スで保存されているので、カイネ-ス活性に重要な役割をはたすと考えられる。2)Ws/Wsラットはマスト細胞欠損、貧血、メラノサイト欠損という点まではWミュ-タントマウスと同じ症状を示すわけだが、Wミュ-タントマウスでは生殖細胞欠損のため、オス・メスともに不妊になるのに対しWs/Wsラットはオス・メスとも生殖力を持つ。この点とWs/Wsラットの貧血が離乳後改善する点は、Ws/Wsラットのマスト細胞欠損ラットとしての有用性を高めるものである。3)Wsミュ-タント遺伝子を種々の近交系に導入する計画は近藤と西村により進められている。近交系のWs/Wsラットが得られれば利用価値は格段に増す。
