研究概要	ラットのWs遺伝子座はc-kitレセプタータイロシンカイネースをコードしている。Ws遺伝子座に小さな欠失を持つWs突然変異遺伝子がホモになったWs/Wsラットはマスト細胞を欠損している。まずWs/Wsマウスのマスト細胞欠損を再度吟味した。いままでマスト細胞を算定するのに、アルシアンブルーで染色を行っていたが、アルシアンブルーで染色されるのはマスト細胞の顆粒中のプロテオグリカンである。マスト細胞の分化過程で顆粒が形成されるに先だって、c-kitレセプターや高親和性IgEレセプターのmRNAが発現される。そこでWs/Wsラットの皮膚におけるc-kitとIgEレセプターβ鎖の発現をしらべたが、Ws/Wsラットの皮膚では正常(+/+)ラットの皮膚の1%以下のmRNAしかみとめられなかった。またin situハイブリダイゼイション法によっても、c-kitレセプターや高親和性IgEレセプターを発現している細胞をみとめることができなかった。すなわち、Ws/Wsマウスの皮膚は、アルシアンブルーで染色される成熟したマスト細胞ばかりでなく、より未分化なマスト細胞も欠損していることがわかった。Ws/Wsラットと+/+ラットに消化管寄生虫Nippostrongylus brasiliensisを感染させると、Ws/Wsラットでも小腸にマスト細胞が出現する。このマスト細胞の表現型をラットマスト細胞性蛋白質分解酵素に対する抗体を用いてしらべたところ、粘膜型であることがわかった。Ws/Wsラット小腸のマスト細胞は数の上では+/+ラットの場合の10%にすぎないが、N.brasiliensis感染により末梢血中の好塩基球は+/+ラットと同様50倍以上に上昇する。N.brasiliensis感染後も皮膚などに結合組織型マスト細胞は出現しないから、分化に対するc-kitレセプターの必要性は、結合組織型マスト細胞、粘膜マスト細胞、好塩基球の順に少くなると考えられる。
