光合成系アンテナの構築原理の解明とその光エネルギー獲得システムへの応用

研究概要

我々は2種の耐熱性緑色光合成細菌Chloroflexus aurantiacusおよびChlorobium tepidumの光合成膜から光合成アンテナを単離、精製した。固体状態での高分解能NMR(野澤担当)を活用し、単離した光合成アンテナの構造をin-situで解析し、ネイティブな状態でのクロロフィルの配位状態、クロロフィルークロロフィル間相互作用、クロロフィルと周囲のアミノ酸残基との相互作用を明らかにした。アンテナタンパク質から光合成アンテナを構成する色素(バクテリオクロロフィルcおよびd)を有機溶媒抽出処理で単離し、HPLCで精製し、これらの色素、およびタンパク質の構造を液体NMR(野澤担当)表面増強IR法(大澤担当)を用いて決定した。さらにアンテナ系における励起エネルギー移動を耐熱性緑色硫黄光合成細菌Chlorobium tepidumのアンテナ系さらにはそこから反応中心への励起エネルギー移動を時間分解ケイ光法で検討を行い、生物が光エネルギー獲得のために構築しているアンテナ系の構造と組立、および集光機構を探った。また、人工光獲得アンテナの構築を構造の明らかとなった色素を用いて行い、その会合特性を光化学特性とをあわせ検討した。これら会合体の構造の知見は固体高分解能NMR(野澤担当)と表面増強ラマン、表面増強IR(大澤担当)で初めて検討可能となった。
