生物認識機能をもつ生体機能材料に関する糖質工学的研究

研究概要	オリゴ糖や多糖の分子認識・細胞認識に基づく精妙な分子生体機能の本質を、分子レベルあるいは細胞レベルで解明し、その基盤に立って新しい生医学材料や生理・薬理活性物質を創出することを目的に、次の糖質高分子物質を合成した。これらの高分子物質のいずれにも、糖鎖が認識信号として濃密に組み込まれているのが特徴である。低分子物質とは異なった特有の動的精密構造認識のいくつかの発現も明らかにした。 1.アンヒドロ糖誘導体の開環重合を経由して3ーデオキシー3ーフルオロー(1→6)ーαーDーグルコピラナンを合成した。デオキシフッ素化した明確な構造をもつ多糖はいままで報告例がない。抗体や酵素への取り込みの解析のためのモデル物質として特異な機能を発揮するのではないか。 2.酵素触媒合成法を活用して高い生理活性をもつオリゴ糖を合成する試みが盛んになっているが、糖鎖高分子合成への応用例はない。糖タンパク質や糖脂質のオリゴ糖成分であり、分化抗原やガン関連抗原として重要なNーアセチルラクトサミン糖鎖に着目して、p-アクリロイルアミノフェニル配糖体、およびpービニルベンジル配糖体の2種類のルートを経て糖鎖高分子を合成した。 3.加リン酸分解酵素の逆反応により、任意の重合度からなるアミロースを酵素触媒合成する手法が開発されている。この方法を応用して、アミロース置換スチレンモノマーを合成し、そのラジカル単独および共重合を行って、多糖を側鎖にもつ構造の明確な高分子を合成した。 4.グルコースの3位をエーテル結合を介してポリスチレンに結合した糖質重合体をコートした培養皿の上で、赤血球細胞が今まで全く知られていなかった特異な挙動をとることを見つけた。グルコース輸送体タンパク質との特異的相互作用によることを突き止めた。 5.アシアロ糖タンパク質レセプターを経由する肝細胞の興味ある組織形成挙動を見出した。
