植物病原糸状菌の病原性遺伝子の単離

研究概要	病原性決定因子として宿主特異的毒素を生産するAlternaria alternata病原菌群について、以下の研究を行った。 1.A.alternata病原菌の病原性遺伝子の単離:A.alternata病原菌群のうち主にナシ黒斑病菌(Japanese pear pathotype、AK毒素生産菌)を用いて、その毒素生産(病原性)遺伝子の単離を試みた。塩基置換を引き起こす突然変異誘発物質を用いて、AK毒素欠損(病原性失活)変異株を分離した。分離した毒素欠損株を親株の染色体DNAコスミドライブラリ-によって形質転換し、毒素回復株を選抜した。約7千株の形質転換体の毒素生産性を検定した結果、毒素回復株は検出されず、残念ながら毒素生産遺伝子の単離には致らなかった。しかし、同様な手法を用いて本菌のメラニン生合成に関与する3つの遺伝子を単離した。黒色色素メラニンは本菌の高い腐性能力に関与し、圃場における広い意味での病原性(寄生適応性)に重要な役割を果たす。単離した遺伝子群の解析から、メラニン生合成に関与する3つの遺伝子が本菌染色体DNAの約40kb領域内にクラスタ-として存在することを明らかにした。また、パルスフィ-ルド電気泳動によって本菌染色体を分画し、メラニン生合成遺伝子クラスタ-の構造をさらに解析するとともに、それぞれの遺伝子の発現様式について調査した。本研究で確立した種々の手法は、毒素生産の遺伝子研究に今後大きく貢献するものと考えられた。 2.宿主特異的毒素の作用機構の解析:A.alternata病原菌群の病原性研究では、宿主特異的毒素の作用機構の解明も重要な課題である。本研究では、イチゴ黒斑病菌(strawberry pathotype)とトマト・アルタナ-リア茎枯病菌(tomato pathotype)のそれぞれAF毒素とAL毒素について解析し、これまで見いだされていなかった新たな作用を明らかにした。
