形質膜裏打ちの超微形態と機能との相関

研究概要	1.骨格筋細胞の形質膜裏打ちとジストロフィン局在モルモット下肢筋について、裏打ち蛋白質であるジストロフィンの局在様式を蛍光抗体染色し、共焦点レーザー顕微鏡にて観察した。ジストロフィン染色は筋形質膜に沿って不連続性に局在した。また、筋線維端には極めて強い染色性が認められた。この部位は電顕的に裏打ち構造が発達していることから、ジストロフィンが筋原線維の膜付着に関与している可能性が示唆された。膜関連蛋白質の一つであるビンキュリンは形質膜に沿って横紋状に規則的な染色性を示し、ジストロフィンとは独立した局在であった。 2.切腱した筋腱連結部の変化ラットヒラメ筋を切腱したのち、経時的に筋腱連結部の形質膜裏打ちの変化を電顕的に観察した。切腱1週で、筋原線維は強く破壊され、筋形質膜との連結を失い、Z板は肥厚したが、裏打ちの肥厚は認められなかった。Z板と裏打ちは互いに性質が異なることが示唆された。 3.ガラス接着マクロファージのアクチン細胞骨格マウス腹腔からマクロファージを採取、カバーグラスに接着させ、接着面における細胞骨格の分布および形質膜との相互関係を電顕的に観察した。接着点に一致してアクチンフィラメントからなる星状構造が形質膜を裏打ちし、ガラス接着を補強していることが明らかになった。 4.腱上皮細胞の形質膜裏打ちと微小管サワガニ歩行脚の横紋筋において、筋収縮力をクチクラ板に伝える腱上皮細胞内には無数の微小管の平行配列が観察され、微小管の両端は裏打ちを介して形質膜に連結されていた。微小管と裏打ちとの間には微細な線維構造が架橋しているのが見られ、裏打ちが微小管の形質膜付着にも力学的役割りを果たしていることが示された。
