移植・培養縫線核ニューロンに対する成長因子としての脳内ペプチドの形態学的研究

研究概要

胎生期ラットからの脳幹縫線核組織を成熟ラット小脳に移植し、セロトニンニューロンが宿主組織に対してどのように再生していくのかを検索した。成熟ラットの脳室に5,6-DHTを投与し、内在性のセロトニン線維を化学的に変成した後に、中脳と延髄の2つの領域からのセロトニンニューロンをそれぞれ小脳へ移植した。中脳由来のセロトニンニューロンは小脳内で正常にくらべてはるかに豊富な突起の伸長(hyperinnervation)を示したのに対し、延髄由来のセロトニンニューロンは正常とほぼ同じ分布パターンを示した。セロトニンニューロンの海馬組織への軸索伸長能力を、セロトニン線維を破壊した成熟ラット海馬への縫線核組織の移植、成熟ラット前眼房水内への縫線核組織組織、縫線核・海馬組織の組織スライス培養法(organotypic slice co-cultures)を用して検討した。いずれの実験においてもセロトニンニューロンは軸索を海馬内に伸長させ、同様の分布パターンが示された。特に密なセロトニン神経叢が歯状回の顆粒細胞の基底部に存在し、この部位にはGFAP陽性、s-100β陽性のグリア細胞が密に分布した。以上の結果から、セロトニン線維とグリア細胞の分布に相関性が示された。グリア細胞の主要構成タンパク質であるs-100βタンパク質を遺伝的に欠くミュータントマウスを用いて、免疫組織化学的(セロトニン、s-100β)にs-100βタンパク質のセロトニンニューロンの発達に及ぼす影響について検討した。その結果、homozygoteの海馬においてs-100β陽性グリア細胞は認められず、さらにセロトニン陽性線維の分布がきわめて少なく、変性セロトニン線維も観察された。このことは、s-100βとセロトニン線維の発達の間に関連性が認められ、セロトニン線維に対するs-100βの神経成長因子としての働きが示唆された。
