病巣指向性酵素誘導体の分子設計と活性酸素病態の制御

研究概要

虚血性心筋障害や脳血管障害をはじめとする各種の疾患において、活性酸素やフリ-ラジカルにより病態が増悪することが知られている。このため、強い酸化的ストレスを伴う病態の分子論的理解とその有効な治療法の開発が重要な課題となっている。酸化的ストレスを軽減するには病巣局所の細胞外空間やそれが低濃度の細胞内区画に抗酸化防御系を動員し、活性酸素種を有効に消去することが重要である。本研究は、化学修飾および遺伝子工学的手法を用い,SODをはじめとする抗酸化防御酵素を特定の組織、細胞およびオルガネラの局所にタ-ゲッティングすることにより、疾患時に見られる活性酸素障害の分子機構を解明すると同時に、その代謝制御の観点から病態改善を目指す新しい治療法確立のための理論基盤を確立することを目的として計画された。
その結果、ヒトCu/Zn型SODを用い,(1)アルブミンと可逆的に結合して極めて長時間血中を循環し,pHが低下した組織病巣部位に濃縮動員されるSOD誘導体(SM‐SOD)、(2)長鎖脂肪酸を共有結合した細胞膜脂質親和性SOD(AC‐SOD),および(3)遺伝子組換えにより血管内皮細胞上のプロテオグリカンに高親和性を示す内皮細胞指向性ハイブリッドSOD(HB‐SOD)のFusion Geneを構築し、その大量発現系を開発することが出来た。さらにSM‐SODが個体レベルでの虚血再循環性不整脈、致死性不整脈、脳乳腫の発現を極めて有効に阻止すること、またAC‐SODが内毒素性角膜潰瘍を著明に軽減することを示してきた。今後これらの酵素誘導体を用いて各種疾患時において、病巣組織細胞の特定の区画でス-パ-オキサイドを特異的に消去することの病態生化学的意義を解析する。
