溶血性連鎖球菌の菌体表面上に存在するM蛋白の抗食作用の分子遺伝子学的解析

研究概要	目的;Streptococcus pyogenesは菌体表層上のM蛋白の抗原性の相違により80種類以上に別れるが、M蛋白をもつ菌は多核白血球の貧食に低抗性を示すことからM蛋白が重要なビルレンス因子と考えられている。我々は、SSPーPCR(Single Specific Primer Polymerase Cham Reaction)法を用いてC203株からemm3遺伝子のクロ-ニングを試みた。方法;Mb構造遺伝子を含んだpJRS42.50(Dr.June R、Scottより供与)から大部分のM6遺伝子を保有したMspI/PvuII fragmentを得、精製した。S.pyogenes C203株由来の遺伝子は菌体をSDSで溶かし、proteinase K.CTAB(hexadecyl trimetyl ammonium bromicle)を用いて蛋白,polysaccharideなどを除き,DNAをisopropanol沈殿により回収した。M6遺伝子由来のMspI/PvuII fragmentをDigoxigennで標識し、hybridizationを行った。SSPーPCR法はPrimer1としてM遺伝子のleader seguence中の保存領域のorigonucreotide(5'>TCG CTT AGA AAA TTA AAA ACA GGA ACG>3')とprimer RV(pumer G)のvectorに相補的なorigonucreotideを用いての増幅を行った。結果と考察;Digoxigenin標識MspI/PvuII fragmentを用いてC203株のEcoRl処理DNAとDotーblot hybridizationを行ってみると,positive spotが検出された。更にSSPーPCR法を用いて約1KbのDNAを中心として5ー6本のDNAを増幅させた標品をDigoxigenin標識MspI/pvuII fragmentを用いてDotーblotを行なって見るとpositive spotが検出され、又、Agarose電気泳動後、Southerm blot hybridizationを行って見ると約1kbのDNAとhybridigeした。この約1kbのDNAがemm3遺伝子由来のものと考えられ、このDNAを精製し、更に現在このDNAのsubcloningを試みている。
