長期アルコ-ル摂取の中止による肝癌発症促進機構とその予防

研究概要	非近交系Long Evans(LEA)ラット及びそれより分離された肝炎、肝癌自然発症ラット(LEC)をアルコ-ル含有液体飼料(Lieber食)で飼育し、肝のアルコ-ル代謝関連酵素であるアルコ-ル脱水素酵素(ADH)、アセトアルデヒド脱水素酵素(ALDH)及びマイクロゾ-ムエタノ-ル酸化系Cytochrome P_<450> llE1の各活性をWistarラットを対照として検討した。【方法及び結果】 6〜8週齢のLEA(n=13)、LEC(n=22)及びWister(n=6)ラットを5%エタノ-ル含有Lieber食(オリエンタル酵母)で飼育した。対照食はLieber食中のエタノ-ルの熱量をショ糖で置換した飼料を用いた。飼育開始2週後までにエタノ-ル食投与群のLEA及びLECラットの大半は、死亡した。その死亡率は、飼育開始1週後でLEA(5/9)、LEC(7/17)、2週後までで、LEA(6/7)、LEC(14/14)であったが、対照食群及びWistarラットは全例生存していた。飼育開始1週及び2週後に生存していたラットから肝臓を摘出し、粗ミクロソ-ム分画を調整、ADH及びALDH活性を測定した。また、各ラット肝マイクロゾ-ム中のCytochrome P_<450> llE1発現量をWestern blot法にて検討した。 LEA及びLECラット肝ASH活性は、Wistarラットと差を認めなかったが、肝ALDH活性は、LEA、LEC両ラットともWistarラットの約1/4に低下していた。活Cytochrome P_<450> llE1は、LEA,LEC及びWistarラット間に差はなく、アルコ-ル投与2週後に投与前の約5倍に誘導された。【考察及び結論】 LEA、LECラットでは、遺伝的に肝ALDH活性が低下しているため5%エタノ-ル含有Lieber食を代謝しきれず死亡したものと推察された。次年度は、肝ALDHアイソザイムの検討やエタノ-ル濃度を下げた実験系での検討が必要と思われる。
