Ex-vivoガストリックチャンバーを用いたラット胃粘液分泌測定法の開発と応用

研究概要	抗ムチンモノクローナル抗体の確立とその性状検討 ELISA法を用いたムチン分析法の開発を最終目標として、モノクローナル抗体(MAb)の開発をこころみ、10種類のMAbを樹立した。このうち9種類はムチンの糖鎖に、1種類はムチンのペプチド鎖に特異性をもっていた。申請期間内にはこれらを用いたELISAによって、ムチンの微量測定法を確立することはできなかったが、いくつかの興味あるMAbの性状の検討を行なった。とくにRGM11,RGM41と命名したMAbはそれぞれラット胃体部表層粘液細胞由来のムチンの糖鎖およびラット胃腺粘液細由来のムチンに特異的であることから、今後胃分泌粘液の由来を特定するときに利用できるものと考えられた。還流液中のムチン測定法の検討チャンバーから回収される還流液中の粘液量に対応する高感度ムチン測定法としては、HPLCを用いた高感度ヘキソサミン測定法を応用した。本法を適用するにあたって還流液中の低分子物質(50Kd以下)を効率よく透析除去する方法を工夫した。 Ex-vivoガストリックチャンバー法による分泌粘液測定法の検討 Ex-vivoガストリックチャンバーを用いて、胃分泌液中の粘液量を経時的に測定する条件を確立した。チャンバーに胃を装着し、生理食塩水を還流することによって、胃内腔に分泌された粘液物質中を回収し、その中のムチン含量を測定する方法を採用した。すでに確立している還流液pHや、potential differenceを同時にモニターし、後者が正常値をとる個体を選んで分析をおこなった。本法を用いて、胃粘液分泌に影響を与えることがわかっているcarbachol投与後の粘液分泌動態を検討したところ、投与量を変えることによって異なる分泌状態を示すことが明らかとなった。
