SFME細胞を用いた造血器腫瘍発がん遺伝子のモニタ-法の確立

研究概要

1.研究目的:本研究は、SerumーFree Mouse Embryo(SFME)細胞がヒトがん組織由来のがん遺伝子によって形質転換され、容易に同系正常マウスに腫瘍を形成する性質を利用して、造血器腫瘍をはじめとする悪性腫瘍における発癌遺伝子のモニタ-法を確立する目的でなされている。
2.SFME細胞の培養条件の確立:SFME細胞の培養にはかなりの熟練を要する。安定して本細胞を供給するためには、培地に超純水を使用する必要があり、添加物(EGFなど)は培地交換時に添加しなければならず、あらかじめ添加した培地を作り置いて使用してはならないことが判明した。さらに、ガラス器具は細胞が付着するために使用しないこと、トリプシンおよびトリプシンインヒビタ-を加えてからの時間はできるだけ短くし、速やかに次のステップに入ること、など注意すべき点が明らかになり、現在では順調に培養できるようになっている。
3.ヒトがん遺伝子による形質転換:ヒト膀胱腫瘍より抽出したHーras遺伝子により形質転換を行いBALB/cマウスに移植し腫瘍を形成した。10^6個/マウスの細胞を移植することにより、腫瘍を形成可能であることが確認された。一旦形質転換した細胞は、血清の存在する培地でも増殖できるようになり、選択は容易である。ただし、長期にわたる維持には無血清が望ましく、早めにクロ-ン化して無血清に移す方が良いとの結論を得た。
4.形質転換した細胞の性状の検討:形質転換細胞中、高頻度に肺に転移する細胞を発見し、継代している。この細胞とPCR法を組み合わせることにより、モニタ-法確立の基礎ができるものと考える。
5.ヒト造血器腫瘍細胞から得たがん遺伝子による形質転換:現在ヒト白血病細胞の細胞増殖動態の解析とともに、白血病細胞からがん遺伝子を抽出している。今後形質転換を図る予定である。
