SFME細胞を用いた造血器腫瘍発がん遺伝子のモニター法の確立

研究概要	1.研究目的:本研究は、SerumーFree Mouse Embryo(SFME)細胞がヒトがん組織由来のがん遺伝子によって形質転換され、容易に同系正常マウスに腫瘍を形成する性質を利用して、造血器腫瘍をはじめとする悪性腫瘍における発癌遺伝子モニター法を確立する目的でなされた。 2.SFME細胞の培養条件の確立:SFME細胞の培養にはかなりの熟練を要するが、培地に超純水を使用し、添加物(EGFなど)は培地交換時に添加すること、ガラス器具は細胞が付着するために使用しないこと、トリプシンおよびトリプシンインヒビターを加えてからの時間はできるだけ短くし、速やかに次のステップに入ること、などを厳守することにより、順調に培養できるようになり、必要に応じて細胞が供給可能になっている。 3.ヒトがん遺伝子による形質転換:SFME細胞に、ヒト膀胱癌T24由来活性型Hーras 1遺伝子とマウスcーmyc を導入して得られた形質転換細胞をBALB/cマウスに移植し腫瘍を形成した。10^6個/マウスの細胞を移植することにより、腫瘍を形成可能であることが確認された。 4.形質転換した細胞の性状の検討:形質転換細胞をマウス皮下に接種し、経時的に各臓器への浸潤を観察したところ高頻度に肺に転移する細胞(r/mHMーSFMEー1)を発見した。この細胞とPCR法を組み合わせて、臓器(今回は肺)に転移した腫瘍細胞の数を算定することを試みた。その結果、マウス一匹の肺内に1×10^4個の腫瘍細胞が存在すれば検出可能であることが明らかになった。 5.ヒト造血器腫瘍への応用:現在ヒト白血病細胞からがん遺伝子を抽出しているが、これを用いてSFME細胞を形質転換し、今回の方法を応用することで治療効果の判定とともに、微量残存腫瘍細胞の量的解析が可能になると考える。
