松果体のRetinopotency(網膜細胞分化能)の解析

研究概要	平成3年度(研究初年度)において、松果体にどのような神経細胞分化能が潜在しているのか、その(神経細胞としての)分化形質をいくつか明らかにすることに成功した。これらの分化形質は、いずれも網膜の神経細胞において発現が見られるものであるので、松果体にRetinopotency(網膜細胞分化能)と呼びうるものが存在するといえよう。今年度は、これらの分化形質のいくつかについて、その発現のメカニズムを明らかにする上で以下のような重要な結果を得た。 1.ラット松果体には神経細胞は見られない。ところが、新生仔ラット松果体を培養すると、多数の神経伝達物質GABA陽性の神経細胞が出現する。GABA細胞の分化は、この培養系で観察される視細胞や内分泌型細胞のように、生理的な濃度のノルエピネフリンの添加による影響を受けない。なぜ、組織の松果体でGABA細胞の分化が見られないのか、さらに検討しなければならない。 2.ラット松果体には神経細胞は見られない。新生仔ラット松果体を培養下に移すと、ごく少数の細胞が神経特異抗原(HPC-1、MAP2)を発現するようになる。ところが、培地のKC1の濃度を脱分極を誘発する濃度に上げると、非常に多数の細胞がこれらの形質を発現するようになる。このことは、神経細胞の分化が別の神経細胞の存在によって誘導されることを示唆しており、神経組織の形成を考える上で、興味ある結果である。 3.トリ松果体には(杆状体タイプの)視細胞がある。胚の松果体を培養すると、これ以外に、少数の錐状体タイプ視細胞が分化する。胚の網膜細胞と同時培養することによって、松果体細胞の中に多数の錐状体視細胞の出現が見られるようになった。この結果をもとに、網膜細胞の培養上清液中に錐状体視細胞の分化を促進する因子の存在を確かめた。
