陽炎ビブリオの病原因子遺伝子発現の調節機構

研究概要	腸炎ビブリオの重要な病原因子である耐熱性溶血毒(TDH)をコ-ドする遺伝子(tdh)の発現を促進する調節因子(VpーToxR)を解析した。VpーToxRをコ-ドする遺伝子(VpーtoxR)はコレラ菌の病原因子発現調節因子(ToxR)の遺伝子と52%の相同性を有しており、推定アミノ酸配列も類似し、特に発現調節に関与すると推定される領域およびtransmembrane領域と考えられる部分では非常に強い売似性が認められた。大腸菌中で、クロ-ン化したtdh遺伝子とVpーtoxR遺伝子を共存させた系で、VpーToxRがtdh遺伝子(tdh1〜tdh4の中で特にtdh2およびtdh4)の発現を促進することを確認した。またtdh2遺伝子について、コ-ドン領域上流144bp付近の塩基配列がVpーToxRによる発現促進において重要な役割を果たしていることが明らかになった。ただし、ゲルシフト法によってVpーToxRの結合能を調べたところ、VpーToxRはコ-ドン領域のすぐ上流(68bpまで)に結合することを示唆する成績が得られ、さらに上流(144bp近付)の塩基配列は、結合したVpーToxRとの間の何らかの相互作用によってtdh2遺伝子の発現促進に関与しているのではないかと考えられた。VpーtoxR遺伝子プロ-ブを作製し、これを用いたハイブリダイゼ-ション試験により、この遺伝子はほとんどの腸炎ビブリオ菌株に存在することを明らかにした。AQ3815株を用いて、VpーtoxR遺伝子を特異的に不活化したisegenic変異株を作製した。この変異株と野生株との比較によって、VpーToxRによるtdh遺伝子の発現促進は、KPブロス中で菌を発育させた場合に特に顕著で、発現促進作用は転写レベル(mRNA)でおこっていることを聖らかにした。
