血液凝固VII因子とIX因子の第1EGF様ドメインに発見された新しい糖鎖の意義

研究概要	昨年度の結果では、合成オリゴ糖は、リン脂質、カルシウム、VII因子存在下、IXa因子によるX因子の活性化反応には、まったく影響しなかった。同様に、リン脂質、カルシウム、組織因子存在下、VIIa因子によるX因子活性化反応に対しても糖鎖の影響は見られなかった。これらのことから、このオリゴ糖が、外因系凝固反応に直接関与している可能性は低いことが示された。またVII因子や、IX因子からオリゴ糖を取り除き、それらの機能解析をする必要があった。そこで、市販のグリコシダーゼを用いて、タンパク質分子から糖鎖を取り除くことを試みたが、これら酵素は、まったく作用しなかった。そこで、ウシVII因子をリジルエンドペプチダーゼで消化した後、DEAE-Sepharoseを用いて、オリゴ糖鎖を含むCla-EGF1ドメインを単離した。Gla-EGF1ドメインは、VIIa-組織因子複合体がペプチド基質であるS-2288を水解する反応を拮抗的に阻害した。さらに、EGF1ドメインの機能を調べる目的でウシIX因子を用いて、Stenfloらの報告に基づきキモトリプシン、V8プロテアーゼ、リジルエンドプロテアーゼによる消化とDEAE-Sepharose,Sephadex G-75,MonoQによるクロマトグラフィーによって、EGF1ドメインを単離した。このIX因子のEGF1ドメインは、IXa因子が、VIII因子存在下で、X因子を活性する反応を拮抗的に阻害した。以上の結果より、EGF1ドメインが、タンパク質分子間の相互認識に重要であることは明白であり、この糖鎖がEGF1ドメインのコンフォメーションの安定化や外因系の代謝、あるいは、生合成の制御に関与している可能性もあり、今後の課題である。
