研究概要

IL-3及びGM-CSFレセプターは各々に固有なα鎖と共通のβ鎖から構成される。高親和性αβレセプターは血球細胞だけではなく線維芽細胞でも増殖シグナルを伝達しうるので血球細胞に特異的な分子は必ずしも必須ではない。またα鎖細胞外ドメインとβ鎖細胞内ドメインからなるキメラレセプターはβ鎖と複合体を形成しシグナルを伝達しうる。即ちβ鎖の細胞内ドメインが多量体を形成し、シグナル伝達能を担うものでありα鎖細胞内ドメインの役割はβ鎖細胞内ドメインの多量体化を促進する。またc-fosやc-jun遺伝子発現とc-myc遺伝子発現及びDNA複製に必要なレセプターβ鎖の細胞内領域が区別され,その細胞内シグナル伝達経路は異なる。前者はShc,Ras,Raf,MAPKが関与しハービマイシンにより阻害されないが後者はチロシンキナーゼ阻害剤に感受性である。PMAとCa^<2+>シグナルに応答し,ヘルパーT細胞におけるGM-CSF発現を制御するNF-CLE0は,その構成成分としてIL-2発現に関与するNF-ATを含む。ヒトJurkat細胞からこの因子を精製し、アミノ酸配列を部分決定しそれに対応する3種のcDNAクローンを単離した。またIL-3プロモーターに結合する新たなZn^<2+>フィンガー転写因子cDNAをクローニングした。Th2サブセット特異的なIL-4とIL-5の発現はcAMPによって促進され、Th1特異的なIL-2は反対に抑制される。IL-4とIL-5プロモーターの活性化に関与するNF-PとNF-IL-5Pを同定し、これらがやはりNF-AT様の性質を持つことを示した。NF-IL-5PはcAMPシグナルにより活性化されるがIL-2のNF-ATは逆に抑制される。これらの知見から各種のNF-AT様モチーフを通して作用する種々のNF-AT様因子が各サイトカインに特有な発現パターンに寄与する。
