多型性を示すグルタミン酸受容体の機能と役割に関する研究

研究概要	グルタミン酸受容体は、NMDA受容体、AMPA/カイニン酸(A/K)受容体、メタボトロピック受容体の3種類の異なった受容体からなる。我々は、NMDA受容体とmGluRのクローニングに、又、相手研究者のSeeburg博士のグループは、A/K受容体とNMDA受容体のクローニングに成功している。その結果、NMDA受容体は、全ての性質を有するNRIと、その性質に多様性を与えるNR2A-2Dの計5種類からなる事、又mGluRは、少なくとも7種類のサブタイプ(mGluR1-mGluR7)からなる事を示し、それぞれの性質、調節、発現部位等を明らかにしてきた。更に、これ等グルタミン酸受容体の多様性の理解の基に、個々のサブユニット或いはサブタイプの高次脳神経機能における役割の解析を進め、両グループで多くの新しい事実を明らかにしてきた。これらの研究成果の中で、1)小脳顆粒細胞は、活性化を受けるとNR2AmRNAが選択的に誘導を受け、この特異的誘導が、本神経細胞の生存と細胞死を調節していること、2)mGluR6は、視覚の伝達において光のコントラストの識別を規定する受容体として機能している事、3)mGluR2は嗅覚の伝達系を調節し、嗅いの識別に重要な役割を果たしている事を明らかにしてきた。本共同研究を進めるにあたっては、協同研究者の桝正幸が9月にHeidelberg大学に滞在し、又中西重忠は9月と10月にそれぞれギリシャとドイツの学会でSeeburg博士に会い、グルタミン酸受容体の遺伝子構造、性質、調節機構等を議論し、多様な本受容体群の機能と調節に関する研究成果の情報交換と受容体クローンの交換を行い、本研究の方向性に関する議論を十分に行った。
