研究概要

我々は大腸菌の染色体分配に関与する遺伝子群を解析するために無核細胞を放出しながら増殖するmuk変異株を多数分離し,muk遺伝子群を同定してきた。単離したmukB遺伝子の塩基配列より演繹したMukB蛋白は177kDaの巨大な分子である。本共同研究では,MukB蛋白を精製し,次のことを明かにした。MukB蛋白は500mMKCl存在下で365kDaの二量体を形成する。MukBの二量体はN端とC端が球状で中央部が棒状の形態をとる。棒状領域にはヒンジ部位があり,ここで折れ曲がる性質がある。MukB蛋白はZnイオン存在下でATPおよびGTP結合能を示し,DNA結合能を持つ。
さらに,mukB欠損変異の示す高温感受性を抑制する変異が新しい遺伝子smbAの変異であること,smbA変異株では一群の細胞質蛋白および膜蛋白の発現が変化することを発見した。またいくつかのsmbA変異株が示す低温感受性はを多コピーで抑制する遺伝子mssAとmssBを見いだした。mssBはATP依存性RNAヘリカーゼの遺伝子deaDと同じことが明かになった。
一方,高温感受性ftsH変異株を解析し,ftsH遺伝子の働きを明かにしてきた。FtsH蛋白は内膜を2回貫通する膜蛋白で,そのC端は細胞質ドメインを形成し,ATP結合配列を持つ。このC端ドメインは真核細胞で見つかった一群のATPアーゼと高い相同性を示す。高温感受性ftsH変異株において,隔壁形成酵素ペニシリン結合蛋白3のC端のプロセッシングの遅れと,β-ラクタマーゼの前駆体の畜積が観察された。FtsH蛋白はこれらの蛋白の局在化の過程に働くことが示唆された。またftsH遺伝子が基質特異的プロテアーゼ活性と関係していることも明かにした。
