研究概要

脳内生物活性物質がimmediate early geneを介して、長期効果をもらたすメカニズムの研究の一端としてステロイドホルモンが培養神経細胞のsurvival rateに与える影響とそのメカニズムについて検討した。glucocorticoidはラット海馬及び扁桃核に対してカイニン酸による毒性を用量依存的に増強するのに対して、estrogenは10-5M以上の高濃度では、神経細胞survaival rateを減少させるものの、10^<-6>それ以下の濃度では、用量依存的に神経細胞の生存率を上昇させる事を認めた。このことは、estrogenが胎生末期からpostnatalにかけての時期に神経細胞の分化・突起の伸張に役割を演じていることを考えれば、naturalである。さらに、我々の実験で、estrogenの全身投与によりラット辺縁系で、c-fosの上昇が見られた。この事はestrogenがAP-1siteをの活性化するという事実を裏付けている。glucocorticoidにはこの様な作用がないか、むしろAP-1siteの活性を抑えており、estrogenとglucocorticoidがAP-1を介すimmediate early gene発現に対して相互拮抗的に働くことが考えられた。estrogen及びglucocorticoidがimmediate early geneの発現に影響を与えた後、次のtarget遺伝子がなんであるかという点に焦点をあて実験を続けるのが、今後の課題である。immediate early geneの発現は、protein kinase Cの活性化、それに続く細胞内Ca^<2+>濃度の上昇と深い関係があると考えられる。海馬培養神経細胞に対してestrogenを投与すると、しばしば細胞内Ca^<2+>濃度の上昇が見られ、そのパターンは、2つに分けられた。その1つは、持続が数10秒以内の、一般の神経細胞伝達物質によるそれと同様なものである。いま1つは、持続が10分以上に及ぶlong-term elevationであった。後者のlong-term elevationは、mRNA阻害剤を加えることによって消失した。
肝組織について見出されたhepatocyto growth factor(HGF)が神経組織の修復にも関与していることが推定されている。また、glucocorticoidが組織障害時に見られるHGFの発現を抑制していることが推定される。大阪大学医学部中村敏一教授より供与された、HGF,c-met c-DNA probeを用いた我々の実験でnothern blotting法により、ラットにカイニン酸を全身投与すると、6時間から48時間にかけて、HGFのmRNAが発現することを認めた。HGFのレセプターであるc-met mRNAについてもカイニン酸投与によりHGF mRNAとほぼ同様な時間的経過の発現が見られた。nothern botting法によってこのmRNA発現に対するestrogenおよびglucocorticoidの影響について現在研究が進行中である。
estrogen受容体が胎生末期から、出産直後の期間において一過性に大脳皮質、小脳などに広く出現し、ここにestrogenが働くことにより神経細胞の突起の伸長、分化ひいては神経回路網の形成に役割を持つことが知られている。そこでneufroblastoma cell lineに遺伝子工学的にetrogen受容体を組み込んだcell lineについても実験を進めている。現段階で、この細胞にestrogenを加えることによってneurobloastoma cell lineが成熟した神経細胞に分化することをみとめた。この細胞について実験が進行中である。
