研究概要

この特別推進研究は、研究計画調書に記載した5項目の研究計画があるが、本年度はそのいずれの項目についても見るべき成果が得られた。以下に主要な研究実績を記す。1.センチニクバエの成虫原基の分化に必須なプロテア-ゼの同定。(1)カテプシンLは、本来リンゾ-ム酵素として知られるシスティンプロテア-ゼで異物の消化に関与すると考えられているが、センチニクバエの成虫原基がエクダイソン存在下成虫構造に分化する過程で、カテプシンL前駆体を細胞外に分泌すること、分泌された酵素を抗体で阻害すると成虫原基の分化が停止することが判明した。(2)種々の阻害剤を使った研究から、成虫原基の分化には上記のカテプシンL前駆体以外にもう一つ、細胞内のプロリルエンドペプチダ-ゼが関与していることが明らかになった。これらの成果は、当初提出した『昆虫は一つの蛋白を多目的に使う』という仮説を支持するものである。2.Sarcophagaレクチン遺伝子の転写因子の解析。
(1)Sarcophagaレクチン遺伝子の5'上流に存在するNF-κB結合配列に特異的に結合する分子量59kDa蛋白の精製を行い、その性質を明らかにした。(2)同じ遺伝子の5'上流及びイントロンに存在するAT-ストレッチに特異的に結合する分子量53kDa蛋白の精製を行い、そのcDNAの単離に成功し全一次構造を決定した。これらの蛋白はいずれもこの生体防御蛋白遺伝子の転写因子と考えられるものである。3.抗菌性蛋白ザ-ペシンBのフラグメント化。ザ-ペシンBを四つのペプタイドに分割し、各々のペプタイドの抗菌性を調べたところ、7番から17番までの11個のアミノ酸よりなるペプタイドがもとのザ-ペシンBと同等の活性を持つことが判明した。
