研究概要

本年度は最終年度に当たるが、計画調書に記載した2項目の研究を中心に研究を推進した。以下にその主要な研究実績を記す。1.ザ-ペシンBのcDNAクローニングと発現:(!)ザ-ペシンBは抗菌活性とカリウムチャンネル阻害活性を併せ持つ蛋白で、本研究の中心テーマの一つである「二重磯能性」を持つ生体防御蜜臼である。ザ-ペシンBのcDNAを単離した結果、ザ-ペシンBはシグナル配列、プロセグメントおよびザ-ペシンB三部分からなる前駆体が合成され、翻訳後のプロセッシングによって成熟蛋白を生じることが判明した。(2)ザ-ペシンBはザ-ペシンと異なり殆どが脂肪体で合成されることか分かった。(3)ザ-ペシンBは正常な幼虫の脳に一を定量存在することか判明した。この事実は、ザ-ペシンBかザ-ペシンとは明らかに異なる挙動を示し、脳内で何らかの機能を発現していることを示している。これらの結果から、ザ-ペシンBの二重機能性が鮮明になりつつある。2.ザ-ペシンBのαヘリクッス部分を改変した新しい抗菌ペプチドの合成:この研究は予想以上に進展しており、現在アミノ酸9残基よりなるペプチドを数種類取得している。これらのペプチドのあるものは、活性か非常に強く、真菌や線虫に対しても有効であった。この研究は現在このペプチドの実用化へ向けて検討を開始した段階にある。3.脂肪体基底膜上の200kDa蛋白結合部位の同定:この2O0kDa蚤白は体液細胞の表面にあり、自己と非自己を識別する働きを持つと考えられている。(1)脂肪体の膜画分に200kDa蚤臼と特異的に結合する活性があることを明らかにした。(2)ついで、この活性を但う蚤白の精製を試みた。その結果、分子量220kDaと230kDaの蛋白の二重体構造を持つ蛋白が精製された。この蛋白はおそらく、200kDa蛋白のリセプターと思われる。
