研究概要	1.RBP-Jκ遺伝子破壊マウスが得られ、このホモマウスの解析を行なったところ、胎生8日前後に死亡することが明らかとなった。特に体節の後部に著しい成長遅延が見られ、また高頻度で神経管の閉鎖不全も見られた。このような結果からRBP-Jκ遺伝子はマウスの発生初期においてきわめて重要な役割をしていることが明らかとなった。一方、ショウジョウバエにおけるRBP-Jkの機能を詳細に解析した結果、感覚母細胞の運命決定とソケット細胞とシャフト細胞の分化決定の少なくとも二段階で末梢神経系の発生を制御することが明らかとなった。RBP-JκはまたE(spl)遺伝子の転写制御に関わり、Hairless遺伝子と競合することが明らかとなった。 2.クラススイッチの分子機構を明らかにするためにin vitroにおけるクラススイッチ誘導細胞株のクローニングを行ない。IL4存在下においてIgAに10パーセント程度の細胞がすみやかにスイッチする系を確立した。この系においてIα領域のメチル化、転写制御がこれまでの報告とは必ずしも一致せず、I領域の転写誘導がクラススイッチ誘導と必ずしも並行しないことを明らかにした。 3.リンパ球におけるprogrammed cell deathの分子機構本年度はsubtraction libraryやdifferential display法により、細胞死やその阻止に際して新たに発現される未知遺伝子を合計5個単離し、現在その機能解析をanti-sense oligo nuculeotideや強制発現法によって検索している。 4.新たにES細胞からストローマ細胞との同時培養により、骨髄系の各種細胞への分化誘導系を確立し、この系を用いてリンパ球の分化制御機構が明らかになるものと期待される。
