研究概要	本年度は、RBP-Jκ遺伝子がマウスBリンパ球の腫瘍化を引き起こすEpshtein Barウイルスの感染に際して、ウイルスゲノムにコードされた腫瘍化に必須の因子EBNA2と会合して多数のEBウイルス、および宿至遺伝子の転写制御に関わることを明らかにした。また、RBPJκはNotchレセプターと直接会合し外来情報伝達を直ちに細胞核に伝え、転写制御に関わることも明らかにした。この結果、RBP-Jκは、神経系および免疫系における細胞の分化と増殖に必須の役割をする情報伝達兼転写制御因子としてのまったく新しい機能分子であることが明らかになった。また、クラススイッチ分子機構の研究において、CH12細胞株はIL-4のみならずTGFBおよびCD40リガンドの三者の混合により高頻度にクラススイッチを引き起こし、この細胞にミニ染色体を導入しin-vitroにおけるクラススイッチ組換えの制御機構の研究を開始するシステムの構築を完了した。細胞死の機構については、細胞死に際して誘導されるCPT-1遺伝子の発現上昇を多数の細胞死関連系において確認し、現在この生理学的意義を検索中である。Bリンパ球細胞株WEHIの抗1gM抗体によって誘発される細胞死がCD40からのシグナルで阻止される機構についても解析を進め、CD40シグナルが細胞周期の制御を行なうことによって細胞死を免れることを明らかにした。
