研究概要

1.家族性大腸腺腫症(FAP)について:FAPの原因遺伝子APCはすでにクローニングされた。本研究班が集積したFAP系の多くでAPC遺伝子の変異が検出され、患者かどうかの遺伝子診断をはじめる事ができた。
FAPは腺腫の数で、密生型と非密生型に分類される。各々の変異を調べたところ、密生型ではコドン1250から1464の間に変異が検出されたのに対し、非密生型の変異はそれ以外の部分にあった。故に、この部位が腺腫形成に強く関係している事がわかった。
FAP患者からの種々の段階の腺腫につきAPCの異常を調べたところ、初期の腺腫に高頻度で両alleleの異常を認めた。この事からAPCの遺伝子の異常は腺腫形成の最初の段階に関与し。不活化の機構は2ヒットである事も明らかとなった。
2.遺伝性非腺腫症性大腸がん(HNPCO)について:HNPCCの原因遺伝子はまだ全くわからず、症例の集積が先決である。HNPCC患者のがん9例を集め、培養樹立例の1例できた。血液は患者20人、家族31人から採血し、EBウィルスを感染させて樹立できたリンパ球は、現在までのところ、患者15例、家族25例である。
HNPCCがん症例のLOHは、染色体17番で3例、18番で3例、22番で0例だった。p53,MCC,DCCについてPCR-SSCPを行ったところ、がん1例にp53の変異が見つかり、MCCで1例の患者のリンパ球に異常が検出され、現在シークェンシングを行っている。
3.大腸がん細胞への染色体1番の導入:導入により扁平になった細胞クローンが得られた。6クローン中4クローンでは1番染色体のうち短碗34〜36領域のみが在存しており、ここにがん抑制遺伝子がある可能性が示唆された。
