研究概要	本年度は3年計画の最終年度にあたる。このことを各班員が意識して研究を行なった。成果は各班員毎に以下にまとめた。西川班員はB細胞初期分化各段階の増殖要求性について検討し、他の組識細胞の分化と異なりB細胞分化では全ての分化段階で増殖能力を失う細胞集団が生成することを明らかにした。この現象の原因については、免疫グロブリン遺伝子の再構成によりトリガーされたp53遺伝子による増殖抑制であることを示唆する結果を得ている。須田班員は造血幹細胞に選択的に発現しているCD34について検討し、ストローマ細胞と造血幹細胞の両方に発現がある場合、造血幹細胞の自己再生能が促進することを明らかにした。菊谷班員は既に作成済みの抗ストローマ細胞モノクローナル抗体が認識する分子の同定を続けているが、遺伝子クローニングには至っていない。宮崎班員は、I型糖尿病を誘発するT細胞クローンの除去にH-2L抗原も関与していることをトランスジェニックマウスを用いて明らかにした。八木田班員はマウスCD2の受容体がCD48であることを明らかにすると共に、これまで作成している抗インテグリンモノクローナル抗体を用いて自己免疫病や移植片拒絶反応を調節することが可能であることを示した。垣生班員は、OVAに対するT細胞受容体を受精卵に導入したトランスジェニックマウスを完成させ、このマウスとこれまでに開発してきたストローマ細胞株を組み合わせることでT細胞の選択過程を試験管内で再現できることを示した。緒方班員は、PCRを利用して未熟胸腺細胞が発現しているkinase,phosphatase遺伝子を数多くクローニングした。中野班員は、ストローマ細胞が発現しているサイトカインをクローニングするためのベクターを開発しこれを用いて分泌される分子を実際にクローニングできることを示した。
