癌関連遺伝子からみた温熱抵抗性癌の細胞持性

研究概要	癌の温熱療法において、その治療効果を異にする癌細胞があることが指摘されてきている。いったい癌細胞の温熱感受性の差異はいかなる理由からであろうか。本研究班は癌細胞の細胞持性を3つのアプローチよりおこなった。1.動物培養細胞に各種活性型癌遺伝子を導入することによって温熱感受性に変化があらわれるのかを調べた結果、cot、mosの2種の遺伝子を導入した細胞が温熱抵抗性になった。このことはこれらの癌遺伝子がヒートショックタンパク質の形質発現誘導に何らかの影響を与えるものと示唆している。2.温熱感受性の異なる癌細胞において、温熱処理後のヒートショックタンパク質と癌抑制遺伝子の一種である野生型p53遺伝子産物の温熱処理による分子安定性を測定した。温熱抵抗性癌細胞(A-7)は44ﾟC、30分間の温熱処理により、約10時間から24時間目ぐらいに野生型p53の蓄積がみられた。しかし温熱感受性癌細胞であるメラノーマ(HMV-1)細胞ではそのような温熱による野生型p53の蓄積がみられなかった。またこれら両細胞とも温熱処理後のヒートショックタンパク質72の形質発現誘導の度合いは同じ程度であった。これらのことより、細胞の温熱感受性の要因に野生型p53の役割が大きいと思われる。また他のメラノーマ細胞のうちでp53の点突然変異タイプの細胞では温熱感受性であった。このような研究成果は温熱療法を行う前に、癌遺伝子・癌抑制遺伝子のタイプを知ることにより、より効率の良い温熱療法による治療効果が期待されるものと考えられる。さらに第3のアプローチとして、実際の温熱療法対象の患者からの癌遺伝子・癌抑制遺伝子のタイプの分析をはじめている。特に治療成績とのそれらのタイプとを対応させていくことを期待している。
