ヒト発がんに於けるオカダ酸発がんプロモーション機構の研究

研究概要

オカダ酸と同様にプロテインホスファターゼ1と2Aの抑制を示すものをオカダ酸クラス化合物と呼ぶが、これらは新しい発癌プロモーション機構を示す発癌プロモーターである。オカダ酸クラス化合物は、いろいろな臓器の発がんを促進した。本年度は、ミクロチスチンによる肝プロモーション機構をヒト肝プロモーションのモデルと考え、研究を行った。DENでイニシェートしたF344雄ラットに、ミクロチスチン(25μg/kg)を週に2回、i.p.で12週間投与したところ、ミクロチスチンは再生肝処置なしで、強力な肝発癌プロモーション活性を誘導することが証明された。また、ミクロチスチンと同様、アオコに含まれるノジュラリンは、5個のアミノ酸からなる環状構造を持つが、この肝発癌プロモーション活性は、ミクロチスチンのそれより強力であり、現在見出された肝発癌プロモーターの中で最強のものと考えられる。ミクロチスチンやノジュラリンを初代培養肝細胞に処理すると、いろいろな蛋白質がリン酸化を受けていた。特に強くリン酸化された2つの蛋白質について、55kDaの蛋白質はサイトケラチン8であり、49kDaの蛋白質はサイトケラチン18であることを証明した。次に、再生肝の細胞核では、オカダ酸の濃度に依存して、Rbとp53のリン酸化は亢進した。更に、ヒト繊維芽細胞でオカダ酸処理によるRb及びp53のリン酸化は増加した。これより、オカダ酸はがん抑制遺伝子産物のリン酸化亢進を誘導していると考えられる。さらに、がん細胞はプロテインホスファターゼが抑制された状況下にあると考えられたが、NIH3T3と形質変換細胞であるF25細胞とでは、プロテインホスファターゼ1と2Aの活性に全く差は認められなかった。これらの活性阻害は、in vitroのレベルで反映できるほど大きいものではないと考えられる。今後、オカダ酸機構のヒト発がんへの応用という考えで研究を進める計画である。
