研究概要	DNA組換え技術を用いて育成された微生物の力によって、人為起源のさまざまな難分解物質を安全かつ迅速に無害化することを目的として、3つの方向から研究を行なった。第一のグループでは未だ分解酵素やその遺伝子の解析が行なわれていない各種の環境汚染物質、すなわちクロ個アニリン(青木)、2,4,6-トリクロロフェノール(浄原)、リグニンモデル化合物(諸星)、ポリエチレングリコール(河合)などの分解系および脱窒関連酵素系(飯島)をとりあげて、それらに関与する分解酵素遺伝子のクローニングと解析を進め、多くの酵素遺伝子について新たな知見を得ることが出来た。第二のグループでは、分解酵素遺伝子についての解析結果を活用して、安全で有用な宿主-ベクター系の構築、より高い分解活性発現能をもつ組換え体の育成方法の確立、基質特異性を拡張のための遺伝子改変を試みた結果、宿主内で安定に保持される安全なベクターpVK100の開発(福田)、γ-BHC分解酵素系の再構築(高木)、1-クロロブタン(1-CB)資化性菌の2種の1-CB分解酵素遺伝子を連結して基質特異性をある程度広げること(児玉)などに成功した。第三のグループでは、育成された組換え微生物を活用するための環境中での動態解析、制御を目指して自然界から取得された分解菌や、分解酵素遺伝子を導入された微生物を対象として、高感度かつ定量性よくモニターする方法としてPCR法とMPN法を組合せた検出法を開発(遠藤)するとともに、組換え菌のマーカーとして安定に保持される水銀耐性遺伝子(mer遺伝子)を用いて組換え体の生残性や環境への影響を調べた結果、非組換え体と差がないこと(矢木)を明らかにした。また、γ-BHC分解菌では基質としてのγ-BHCを土壌孔隙中に十分拡散させた場合に土壌に定着出来ること(松本)が確認された。以上の研究班全体の成果に基づき、少なくとも閉鎖系での環境浄化へ向けて実用化の可能性が示された。
