研究概要	耐熱性酵は単に熱安定性に優れているだけではなく、プロテアーゼによる分解などの生物的な作用も受けにくいことから、センサーやバイオリアクターにおける安定な機能素子として利用されている。われわれは、基質特異性は異なるが同種の反応を触媒する、アラニン、ロイシン、フェニルアラニンの各脱水素刻素を種々の好熱菌より単離し、それぞれの一次構造を決定するとともに、これまでに立体構造が決定された各種のNAD(P)依存脱水素酵素との構造比較から、これら3種のアミノ素脱水素酵素には、NAD(P)を結合し、ヒドリドイオンを転移し、酸化還元反応を行う場である触媒性ドメインと、アミノ酸の構造を認識し、これを結合して反応を行う基質結合ドメインの2つの部分から構成されることを示した。このうち後者については、基質認識を行っているいわば調節性のドメインとして機能していることを明らかにした。耐熱性酵素の各ドメインは常温菌由来の酵素のそれに比べて機能的、構造的に強い狭立性を持っていること、さらに、耐熱性酵素においては、各ドメインの独立性が高いため、調節性ドメインに何らかの修飾を加えてもヒドリドイオンの転移という酵素の触媒作用には障害を与えないことを想定し、耐熱性ロイシン脱水素酵素の機能性ドメインにフェニルアラニン脱水素酵の調節性ドメインを結合したキメラタンパク質を作成し、ロイシン脱水素酵素からフェニルアラニン脱水素酵素への変換の有無を検討し、このドメインの構造と機能と関連を検討した。これら一連の研究によって、耐熱声酵素の各ドメインをタンパク質工学の手法を用いて改変し、入れ換えることによって、複合的な機能を持つ素子としての新しい耐熱性酵素を作出できることをはじめて実証した。
