研究概要	本研究班では、核酸の構造や代謝の分子論的な解析を基にした機能性ヌクレオシド誘導体の創生を計ることを目指している. 松田は、糖部や塩基部を化学修飾したヌクレオシドをDNAオリゴマーに化学的に導入したとき機能性分子として作用する以下の新しい誘導体の設計と合成を行なった.1)RNAとのみ二本鎖を形成するアンチセンス核酸として、糖部1^1-位にアミノリンカー導入したデオキシウリジン誘導体を合成し、DNAオリゴマーに導入したとところ予想したとうりRNAとのみ安定な二本鎖を形成することが分かった.さらに、このオリゴマーはヌクレアーゼに抵抗性を示した.2)グリコシル結合回りのコンフォメーションをアンチ型に固定した新しいヌクレオシド誘導体、3-アルキル-3-デアザイノシンの合成に成功した.今後、核酸中への導入を行ない核酸の構造変化への影響を検討する.牧は、光励起下で活性酸素の模倣物として機能する特異な複素環N-オキシド(POP)を用いてヌクレオシド類の光酸化反応を検討し、ヌクレオシドラジカル種が発生しているという化学的な傍証を得た.さらに、POPがDNA鎖を効率的に切断することを明かにした.宮坂は、HIV-1の逆転写酵素(RT)に特異的に結合する光標識剤を合成した.この化合物は、リード化合物であるHEPTの約640倍の抗HIV-1活性を示し、X-線結晶解析によりそのコンフォメーションも明かにした.今後RTに対する光標識を行ない結合部位を明にする.綿矢は、マラリア原虫の核酸代謝、特にピリミジン代謝が動物のそれと異なることに着目し、5-fluoroorotate(FO)に強い抗マラリア作用を見いだした.FOは、予想どうり動物細胞の増殖を抑えず、高い選択毒性を示した.
