研究概要

シンク細胞に有用物質が大量に集積する場合には、細胞オルガネラが物質の貯蔵庫としての機能を持つように変化分化する。例えば、プラスチドはデンプンを貯蔵するアミロプラストに、液胞は貯蔵タンパク質貯蔵型のプロテインボデイーに転換する。アミロプラストおいては、プラスチド遺伝子の発現が葉緑体の場合の62%の抑えられていた。液胞とプロテインボデイーの間の相互転換に際しては液胞膜の融合・出芽が起こるが、これに関与するカボチャの膜タンパク質のcDNAをクローニングし、このタンパク質の構造を解析した。また、液胞の形態形成・維持に関与する遺伝子を酵母から単離し、その構造を明らかにした。さらに、高等植物液胞膜に存在するH^+ーATPaseおよびピロホスファターゼの構造を明らかにし、液胞膜を通じての物質輸送にこれらの酵素が重要な役割を演じていることを提示した。特に果実の場合には、液胞膜H^+-ATPaseが他の植物のものとその性質を異にしていて、有機酸や糖の集積と密接な関係を持っていることを示唆した。一方、プロテインボイーにおける貯蔵タンパク質の集積機構を明らかにするために、貯蔵タンパク質のcDNAの解析および改変を行い、貯蔵タンパク質の構造がその大量集積と深い関わりあいをもっていることを明らかにした。すなわち、貯蔵タンパク質のプロセシングおよびジスルフイド結合の形成が分子集合、ひいては大量集積に必須であることを明らかにした。また、デンプンの集積の分子機構を明らかにする目的で、ジャガイモ塊茎アミロプラスト中の糖代謝酵素のcDNAを解析中である。さらに、カタラーゼ遺伝子の5'上流域を用いのトランスジェニック植物の作出を通じて、この5'上流域中に種子中で遺伝子が強力に発現することに関与しているシスエレメントが存在していることを明らかにし、その存在領域を示唆した。
