研究概要	我々は、チロシンキナーゼの生理機能を明らかにすることが細胞がん化の機構を解明する上で重要であると考えて以下の研究を行なった。 1.lyn遺伝子を欠損する細胞株等に遺伝子を導入する実験から、Bリンパ球の抗原受容体と会合するLynキナーゼがSykキナーゼと共存することにより、細胞内蛋白質のチロシンリン酸化反応を促進することを示した。又fynを過剰発現するT細胞ハイブリドーマでは、Fynの活性化がZAP-70キナーゼの活性制御に関わっていることを示唆する結果を得た。一方、CD30陽性リンパ腫でチロシンリン酸化が亢進しているp80蛋白質を同定し、その分子生物学的解析を進めると共に、リンパ腫発症に於けるp80の意義を検討している。 2.脳で発現する新規チロシンキナーゼ遺伝子fakとbrtのcDNAをクローニングした。fakは同時期に外国のグループによって細胞接着に関わることが示されたが、我々はfakの脳特異的産物を見いだしており、fak欠損マウスの作成や、免疫生化学的手法を用いてその機能解析を進めている。又Fynがミエリン形成に関わることを示し、更にfyn欠損マウスでミエリン形成が不完全であることを示した。 3.受容体型チロシンキナーゼErbB-2の標的蛋白質としてTob-1ならびにTob-2を同定し、そのcDNAクローニングを行なった。tob-1遺伝子はヒト染色体17q22にマップされること、そしてTob-1蛋白質が細胞増殖抑制活性を有することを示した。又、エストロゲンがErbB-2チロシンキナーゼ活性を促進するという昨年度の成果に基づいて、エストロゲンに抗がん剤を結合させ、ErbB-2過剰発現細胞を選択的に殺傷することに成功した。
