研究概要	1. EGF受容体のシグナル伝達の解析 EGF受容体からのシグナル伝達に関する現在のモデルは、カルボキシル領域におけるチロシン自己リン酸化の重要性を強調したものであるが、他の機構がないかは不明である。我々は、カリボキシル領域の種々の変異体を作成し、GAPやPLCγの結合がもはや起こらない自己リン酸化部位欠損型EGF受容体の変異体(DEL1011+F992)でも、DNA合成や細胞のトランスフォーメーション能はまだまだ保たれていることを示した。非常に興味あることには、このEGF受容体変異体の場合でも、Shc自体のチロシンリン酸化、Grb2との会合、Rasヌクレオチド交換因子Sosの“活性化"(リン酸化による移動度の変化)が認められることである。これらの点は、従来のモデルからは予想できなかった点であり、EGF受容体からのシグナル伝達にalternative pathwayが存在すること、その際Shcの活性化が重要な位置を占めることを、強く示唆している。 2.内皮細胞特異的シグナル伝達系 VEGF-Flt受容体の解析我々が初めて単離したFlt受容体のmRNAがラット肝においては類洞壁内皮細胞画分に強く発現し、またVEGFがこの細胞画分(以下NP-cell)に対して強い増殖刺激活性をもつことを示した。この点をさらに解析し、VEGFの効果が極めて特異的であること、すなわち、従来内皮細胞増殖因子の1つとして用いられてきた塩基性、酸性FGFが全く増殖刺激活性を示さない興味ある系であることを見いだした。さらに、ヘパリンがVEGFの効果を増強することも確認した。Flt受容体のシグナル伝達系解析のモデル系として、NIH3T3線維芽細胞をもちい、リガンドの結合や細胞内でどのようなシグナル伝達系が働いているかを解析し、それが特異的な性格をもつことを明かにした。
