研究概要	(1)骨髄異形成症候群(MDS)・白血病とras遺伝子:MDS細胞や白血病細胞に見い出されるras遺伝子の変異は多くの場合、N-ras遺伝子の変異で、12、13、61番目のいずれかのアミノ酸コドンにおける点突然変異である。MDS細胞や白血病細胞では15〜20%の頻度で見い出される。MDSや白血病におけるras遺伝子の変異は、N-ras遺伝子の変異が多く検出される。ras遺伝子の変異を解析することは、MDSの白血化に関する予後判定に有用である。 (2)MDS・白血病とp53遺伝子:p53遺伝子は癌抑制遺伝子であり、両方のアレルで欠失、変異、挿入などを生じてp53遺伝子が不活化されることによって細胞を癌化に導く。p53遺伝子の変異はMDS症例、白血病症例、白血病細胞株と細胞の増殖能が高くなるほど、変異頻度も高くなること、およびMDSにおいても病期が進行した症例に変異が認められていることなどから、p53蛋白の不活化が細胞増殖に有利な条件を与えることが示唆される。 (3)Ph^1陽性ALL:Ph^1陽性ALLはALLの15〜20%に認められるが、この場合にはPh^1染色体は2種類に大別される。1つのタイプはCMLと同様であり、bcr内に切断点があって8.7kbのmRNAを転写し、p210の蛋白に翻訳される。もう1つのタイプは切断点がbcr遺伝子の第1イントロンにあり、7kbのmRNAに転写されp190の蛋白に翻訳される。Ph^1陽性ALLにおいては両方のタイプの組み換えがほぼ同数例に観察される。 (4)CML急性転化:CML急性転化時に出現する付加的染色体異常の中でt(3;21)(q26;q22)は相互転座であることが特異的である。t(3;21)転座の結果、AML1/EVI-1キメラ遺伝子が形成され、これは融合mRNAへと転写される。このAML1/EVI-1 fusionの形成はRT-PCRで検討したt(3;21)転座を有する3症例で共通であった。t(3;21)転座の結果、キメラ型転写因子が形成され、これがCML急性転化に関与している可能性がある。
