新しいG1,G2期特異的細胞周期阻害剤の探索とそれを用いた増殖制御機構の解析

研究概要	(a)トリコスタチンAとラディシコールによるヒト腫瘍細胞の形態変化と誘導される蛋白質の解析我々はこれまで細胞周期阻害剤トリコスタチンA(TSA)の標的分子がヒストンデアセチラーゼであることを明らかにしてきた。本年度はヒト由来腫瘍細胞HeLa,T24に対する作用を解析し、両細胞ともアクチンストレスファイバーの再形成を伴う著しい形態変化を示すことがわかった。ヒストンアセチル化はクロマチンの転写活性を正に調節するものと考えられることから、TSA処理した細胞中で高度に発現が誘導される蛋白質を探索したところ、アクチン調節蛋白質の一つであるgelsolinがHeLaで18倍、T24では8倍高発現されていることを見いだした。また、チロシンキナーゼ阻害剤であラディシコールにおいても同様の形態正常化とgelsolinの発現誘導が見られた。 (b)分裂酵母のレプトマイシンB耐性遺伝子の解析レプトマイシンB(LMB)の標的分子の同定を目指し、分裂酵母のLMB耐性耐性遺伝子のクローン化を行なったところ、染色体高次構造維持に関わるcrm1の変異遺伝子を取得した。オリジナルなcrm1変異株とLMB処理した野生株との比較を行なったところ、crm1変異株はLMB感受性異常を示すだけでなく、全ての表現形が類似していたことからLMBの標的分子はcrm1遺伝子産物であると考えられた。crm1遺伝子の機能を明らかにするため、crm1変異株のLMB感受性を抑圧する変異を取得したところ、crm1変異と共存するときのみに超耐性を与えるclr1変異を得た。 (c)フローサイトメトリーを用いた新規細胞周期阻害剤の探索 G1、G2期の進行を特異的に阻害する微生物代謝産物をスクリーニングしたところ、放線菌の培養液より新規物質レプトールスタチンを単離・構造決定した。
